Development and standardization of chemical and safety pharmacological experiments using human induced pluripotent stem cells (hiPSC) contribute not only to the facilitation of central nervous system (CNS) disease-related drug discovery, but also to the early detection of adverse drug reactions. In this project, we will employ standard human neurons cultured in multi-well plates and state-of-art immunocytochemical imaging technology to develop a high-throughput assay of “Synaptotoxicity”. Glutamate-dependent drebrin exodus, which is prerequisite for learning and memory, is used as a biomarker of mature synapses. We will standardize the biomarker and phenotype of mature neurons, and validate in-vitro CNS function assessment in a multicenter evaluation. In the future, it is expected for this project to produce the data which will become the scientific basis for synapse toxicity.
ヒトの中枢神経系への医薬品の副作用を予測・評価するためには、記憶・学習をつかさどるシナプス機能への毒性(Synaptotoxicity)を評価できるインビトロ実験系を開発することが必要です。本研究では、ヒトiPS細胞由来神経細胞を利用したハイスループットインビトロ脳機能アッセイ法(HIBA)を開発し、医薬品の神経毒性評価への応用を目指します。
神経細胞の成熟に伴い樹状突起スパイン内へドレブリンが集積することは神経細胞の成熟度のバイオマーカーとなり、グルタミン酸刺激によるドレブリンの樹状突起への移動はシナプスの記憶・学習機能を評価するバイオマーカーとなります。神経毒性評価としてはこれまでは、神経細胞死の評価やてんかん波の発生をマーカーにすることが一般的でしたが、本研究では、高次脳機能への障害を評価するためにドレブリンクラスター数の減少をマーカーとし、多施設間バリデーションを行い、将来的にHIBAの国際標準化を目指します。
本プロジェクトは全国から12大学(20部局)、4研究所、4企業からの研究協力者を招いて、オールジャパン体制で遂行していきます。
